uPA is a serine protease with an extremely limited substrate specificity, cleaving the sequence Cys-Pro-Gly-Arg560- Val561-Val-Gly-Gly-Cys in plasminogen to form plasmin. uPA is a potent marker of invasion and metastasis in a variety of human cancers associated with breast, stomach, colon, bladder, ovary, brain and endometrium. It contains Ser21-Leu431, which consists of two chains: Long chain A (Ser21-Phe177) and chain B (Ile179-Leu 431). The long chain A is further cleaved to yield a short chain A (Lys156-Phe 177) and N-terminal fragment (Ser21-Lys155). Molecular Weight The protein has a predicted MW of 47.5 kDa. Due to protein lysis and glycosylation, the protein migrates to 24-26 kDa(long chain A), 35-38 kDa(chain B) and 52-60 kDa(long chain A&chain B) based on Tris-Bis PAGE result. Physical Appearance Sterile Filtered White lyophilized (freeze-dried) powder. Purity > 95% as determined by SDS-PAGE and HPLC Bioactivity Measured by its ability to cleave a peptide substrate, N-carbobenzyloxy-Gly-Gly-Arg-7-amido- 4-methylcoumarin (Z-GGR-AMC). The specific activity is > 2000 pmol/min/μg Endotoxin < 1.0 EU per 1μg of the protein by the LAL method. Formulation Lyophilized from 0.22μm filtered solution in 25mM HEPES, 150mM NaCl (pH 7.5). Normally 8% trehalose is added as protectant before lyophilization. Reconstitution Centrifuge tubes before opening. Reconstituting to a concentration more than 100 μg/mL` is recommended. Dissolve the lyophilized protein in distilled water. Storage The products are shipped with ice pack and can be stored at -20℃ for 1 year. 2-7 days, 2 to 8 °C under sterile conditions after reconstitution.3 -6 months, -20 to -80°C under sterile conditions after reconstitution.
